EVALUASI HISTOPATOLOGI ORGAN TESTIS TIKUS JANTAN (Rattus norvegicus) GALUR WISTAR SETELAH PEMBERIAN EKSTRAK ETANOL KULIT BATANG PAKAN BANYU (Croton argyratus Blume.) (Oleh : Syarifah Siti Sarah; Pembimbing : Nurlely, Noor Cahaya ; 2021; 35 Halaman)

Pakan Banyu (Croton argyratus Blume.) merupakan tumbuhan berkhasiat kontrasepsi alami di Desa Riam Andungan Kintap, yang diketahui mengandung saponin, alkaloid, tannin dan steroid. Tujuan penelitian ini untuk mengevaluasi gambaran makroskopis organ testis tikus (berat) dan histopatologi mikroskopis organ testis (Tubulus seminiferus) setelah pemberian ekstrak etanol 70% kulit batang C. argyratus. Penelitian menggunakan tikus jantan Galur Wistar yang dibagi menjadi 5 kelompok yaitu kelompok kontrol (larutan Na-CMC 0,5%) dan kelompok perlakuan yang diberi ekstrak dengan dosis 500, 5000, 15000 dan 20000 mg/KgBB. Pengamatan dilakukan selama 14 hari dan pada hari ke-15 testis diambil untuk dibuat preparat histopatologi dengan metode pewarnaan Hematoxylin-Eosin (HE). Hasil yang diperoleh menunjukkan antar tikus kontrol dan tikus yang diberikan ekstrak kulit batang C. argyratus memberikan pengaruh terhadap berat testis dan testis berwarna putih pucat. Gambaran histopatologi testis tikus galur wistar setelah diberikan ekstrak kulit batang C. argyratus mengalami perubahan berupa adanya degenerasi, nekrosis, dan atropi. Perubahan semakin meningkat seiring dengan meningkatnya dosis pemberian ekstrak. Kesimpulan yang didapatkan adalah pemberian ekstrak kulit batang C. argyratus dengan dosis 500, 5000, 15000 dan 20000 mg/KgBB menimbulkan kerusakan pada histopatologi testis tikus.

Pakan Banyu (Croton argyratus Blume.) is a plant with natural contraception in Riam Andungan Kintap, to contain saponins, alkaloids, tannins and steroids. The purpose of this study was to evaluation of testis macroscopic image of male rats (weight) and the microscopic of testis histopathological images (Tubulus seminiferus) between the 70% ethanol extract. The Wistar strain male rats divided into 5 groups, the control group (0.5% Na-CMC solution) and the treatment group given extracts at doses of 500, 5000, 15000 and 20000 mg / KgBB. Observation up to 14 days and on the 15th and the testis were taken to make histopathological preparations using the Hematoxylin-Eosin (HE) staining method. The results showed that the weight and color of the testes between the control rats and the rats that were given C. argyratus bark extract had a significant effect on the weight of the testes and the testes were pale white. The testis histopathological picture of wistar rats after induced C. argyratus stem bark extract experienced changes in the form of degeneration, necrosis, and atrophy. The changes increase with increasing dosage. Therefore, it can be ignored that the administration of C. argyratus stem bark extract caused damage to the testicular histopathology of rats.
