Background: Tooth extraction is the most commonly performed procedure in the field of dentistry, post tooth extraction will result in a wound called a tooth socket. After tooth extraction there are several wound healing processes ranging from haemostasis, inflammatory phase, proliferation phase, and remodelling phase. Bone graft is a material to restore the function of a lost or damaged bone tissue. One of the bone graft materials is allograft from DDM. DDM has osteoconductive and osteoinductive properties so that it can accelerate the bone healing process. Purpose: To determine whether there is an effect of using Demineralized Dentin Matrix (DDM) on the number of osteoblasts and osteoclasts in the bone healing process after guinea pig (Cavia cobaya) tooth extraction on days 7, 14 and 21. Methods: this research is a pure experimental with a posttest with control group design divided into 6 groups. treatment group and control group using a sample of 18 guinea pigs. Results: the results showed that the use of DDM had an effect on the number of osteoblasts and osteoclasts compared to the control group which was only given PEG. One Way Anova test in each group showed that there was a significant difference with p=0.000. Kruskal Wallis test for osteoclast cells obtained a significance result of p=0.006 which means p<0.05 indicates a significant difference between the treatment group and the control group. Conclusion: DDM is proven to be able to affect the increase in the number of osteoblast cells on days 7, 14, 21 and decrease the number of osteoclast cells from day 7, 12 to gradually decrease on day 21.
