ABSTRAK

Penelitian yang berjudul Preservasi Semen Kambing Peranakan Boerbertujuan menguji kemampuan pengencer tris yang dikombinasikan dengan ekstrak daun kelor pada konsentrasi berbeda terhadap kualitas semen kambing peranakan boer yang dipreservasi pada suhu 3-5oC. Penampungan semen dilakukan dengan menggunakan vagina buatan. Metode penelitian ini menggunakan Rancangan Acak Lengkap (RAL), dengan empat perlakuan dan empat kali ulangan yang terdiri dari: 73% pengencer tris + 20% kuning telur + 7% gliserol tanpa ekstrak daun kelor(P1), 71% pengencer tris + 20% kuning telur + 7% gliserol + 2% ekstrak daun kelor(P2), 69% pengencer tris + 20% kuning telur + 7% gliserol + 4% ekstrak daun kelor(P3), 67% pengencer tris + 20% kuning telur + 7% gliserol + 6% ekstrak daun kelor (P4). Setiap perlakuan dievaluasi kualitas spermatozoanya setiap hari hingga presentase motil minimal 40%. Hasil penelitian pada karakteristik semen segar kambing peranakan boer yaitu volume 0,72 ml, konsentrasi 4.036 juta/ml, motilitas 76,00%, hidup spermatozoa 89,00%, dan abnormalitas spermatozoa 5,00%. Hasil penelitian persentase motilitas dan spermatozoa hidup pada hari ketiga menunjukkan semua perlakuan tidak berbeda nyata P1 (45,00%; 54,79%), P2 (43,75%; 57,31%), P3 (42,50%; 61,02%) dan P4 (41,25%; 52,55%). Hasil menunjukkan bahwa pengencer tris yang dikombinasikan dengan berbagai konsentrasiekstrak daun kelor tidak berbeda nyata (P>0,05) terhadap persentase motilitas dan spermatozoa hidup dihari ke-3. Dapat disimpulkan bahwa semua perlakuan dapat mempertahankan kualitas spermatozoa kambing peranakan boer selama tiga hari preservasi pada suhu 3-5oC sesuai ketentuan SNI 4869.3:2014. Konsentrasi antara pengencer tris 69% yang dikombinasikan dengan 4% ekstrak daun kelormenghasilkan presentase motilitas dan presentase hidup spermatozoa kambing peranakan boer lebih baik maka perlakuan P3 yang memenuhi syarat untuk program inseminasi buatan.

ABSTRACT

Study of Preservation of Boer Crossbred Goat’s Semen was to test the ability of tris diluent combined with Moringa leaf extract at different concentrations to the semen quality of Boer crossbred goat preserved at 3-5oC. Semen storage is carried out using an artificial vagina. This research method used a completely randomized design (CRD), with four treatments and four replications consisting of: 73% tris diluent + 20% egg yolk + 7% glycerol without moringa leaf extract (P1), 71% tris diluent + 20% egg yolk + 7% glycerol + 2% moringa leaf extract (P2), 69% tris diluent + 20% egg yolk + 7% glycerol + 4% moringa leaf extract (P3), 67% tris diluent + 20% egg yolk + 7 % glycerol + 6% Moringa leaf extract (P4). Each treatment was evaluated for the quality of the spermatozoa every day until the motile percentage was at least 40%. The results of the research on the characteristics of fresh semen from Boer crossbred goats were volume 0,72 ml, concentration 4.036 million/ml, motility 76,00%, live spermatozoa 89,00%, and spermatozoa abnormalities 5,00%. The results of the study on the percentage of motility and live spermatozoa on the third day showed that all treatments were not significantly different from P1 (45,00%; 54,79%), P2 (43,75%; 57,31%), P3 (42,50%; 61,02%) and P4 (41,25%; 52,55%). The results showed that tris diluent combined with various concentrations of Moringa leaf extract was not significantly different (P> 0.05) on the percentage of motility and live spermatozoa on the 3rd day. It can be concluded that all treatments can maintain the spermatozoa quality of Boer breed goat for three days of preservation at a temperature of 3-5oC in accordance with the provisions of SNI 4869.3: 2014. The concentration between 69% tris diluent combined with 4% moringa leaf extract resulted in a better percentage of motility and survival of Boer crossbred goat spermatozoa, then the P3 treatment was eligible for the artificial insemination program.
